New Construction Pex: Should I Use 2 Manifolds?

I'm trying to figure out how I'm going to configure Pex plumbing in my new build (~5500 SF 2 story excluding garage, 5.5 baths (1.5 first floor, 4 second floor), laundry room (1st floor), kitchen(1st floor)

I know one of the arguments against a pex manifold versus valves at every fixture is that you have to travel to the manifold to shut off the fixture and that it costs just as much as copper then since you have to use home runs from the manifold.

How about 2 manifolds. 1 in the basement for the basement / first floor and 1 on the 2nd floor to cover the second floor? I think the costs will be the same as 1 manifold since the cost of less pex from manifold on 2nd floor will cancel out the cost of the 2nd manifold.

What say you people of plumbing wisdom?
