Kitchen drain has air trapped in drain pipe

My kitchen drain will back up after running for several minutes or also if I fill up the sink and then open the drain. It starts to go then stops and starts getting bubbles coming up through the water. I installed a clean out plug that is outside about 2 feet from the sink. The 2" line goes around the house to the back yard septic tank. I think it joins in the main line somewhere.

If I open the clean out plug, a big rush of air comes out and then it drain fine with the plug out. Why is this happening? I installed a vent under the sink with a studor valve but this didn't help at all because I have pressure in the line and the valve does not let any positive pressure out, only air in. The sink used to drain ok before but now it doesn't. There is a big cast iron stack in the basement that the toilet sits on, at the top there is a hub for the tub and bathroom sink and a 2" vent line is hooked into the same line. Could the vent at the top of the stack be plugged?
