Closing of AC vents in one room

Hi..

I live in a Cape Cod house and we had central AC put in several years ago. The trunk line runs from our basement to the 1st and second floors through pre-existing closet space. To cool the 2nd floor, it goes up into the small attic and duct work branches out to two bedrooms and a bathroom. My son's bedroom is upstairs and the other bedroom is an unused work room that I can close off.

The upstairs gets quite a bit hotter than the 1st floor. I was wondering if I closed the vents in the ceiling to the unused work room if it would send more cool air into my son's room? Or, should I aim to cool the upstairs evenly?
