Help with a new garden

I have a new house on one, sloped acre. The top half I keep mowed, the bottom half gets cut once per year. In between is a 'no man's land' where we pushed all of the rocks that were on the property when it was cleared. There is also a fair amount of dirt there, as it is now overgrown with weeds. I'd like to make this into a wildflower garden. Several questions I would appreciate input on:

1. Can I kill everything there now with Roundup or something comparable?

2. What kind of bulbs/perennials would do well in a sunny, well-drained, west facing, slope of large rocks and dirt?

3. Can this be done this fall?
